Studies of systematic evolution and karyotypic variation in Smilax and Heterosmilax (Smilacaceae)

Smilacaceae are a taxonomically confused, cosmopolitan family of Liliales characterized by their climbing habit, reticulate leaf venation, paired petiolar tendrils, unisexual flowers, and superior ovaries. Deviations from this generalized morphology have led the division of Smilacaceae into at least three different genera and five sections within the large genus Smilax. The karyotypes and chromosomes of 17 taxa of Smilax and Heterosmilax were examined and compared with previous studies of Smilacaceae. Phylogeny and karyotype evolution of the two genera are discussed in this paper along with evolutionary trends in inflorescence morphology, in the context of gene trees based on nrDNA ITS sequences. Karyotypes of these 17 taxa are characterized as follows: (1) “woody S. riparia”, 2n=32=L(2m+2st+2t)+M(4sm+4st)+S(2m+8sm+8st); (2) S. scobinicaulis, from Guizhou, 2n=32=L(2sm+4st+2t)+M(6sm(SAT)+2st)+S(2m+6sm+8st); (3) S. china in Shennongjia, Hubei is a diploid, 2n=32=L(6st)+M(4sm+6st)+S(2m+6sm+8st); (4) S. china in Guangxi with 4x, 2n=64=L(2sm+12st+2t)+M(4sm+8st)+S(6m+20sm+10st); (5) S. china from Japan with 4x chromosomes, 2n=64=L(4sm+14st+2t)+M(6st)+S(4m+28sm+6st); (6) S. trinervula from Guiyang, 2n=32=L(8st)+M(2sm+2st)+S(2m+6sm+12st); (7) S. chingii, 2n=32=L(2sm +6st)+M(4sm+4st)+S(2m+10sm+4st); (8) S. ferox from Kunming, a hexaploid, 2n=96= L(6sm+22st+6t)+M(2sm+10st)+S(6m+28sm+16st); (9) S. lebrunii from Sichuan, a hexaploid, 2n=96=L(4sm+24st)+M(4sm+10st)+S(10m+26sm+18st); (10) S. stans from Chongqing, 2n=32=L(4st+2t)+M(2sm+4st)+S(12sm+8st); (11) S. austro-zhejiangensis found in Zhejiang, 2n=32=L(4st+2t)+M(2sm+6st)+S(12sm+6st); (12) S. umbrosa found in Mt. Emei, 2n=32= L(2sm+6st)+M(2m+2sm+2st)+S(4m+8sm+6s); (13) S. bockii from Mt. Jingfo, 2n=32= L(2sm+6st)+M(6st+2m)+S(8sm+8st); (14) S. guiyangensis from Guiyang, 2n=32=L(6sm+ 2st)+M(4sm+2st)+S(2m+10sm+6st); (15) S. vanchingshanensis from Guizhou, 2n=32 =L(2sm+6st+2t)+M(4st)+S(4m+6sm+8st); (16) S. lunglingensis from Kunming, 2n=32= L(4st)+M(4sm+4st)+S(6m+6sm+6st); and (17) S. polycephala from Yunnan, 2n=32= L(2sm+ 6st)+M(2sm+6st)+S(6m+6sm+4st). The results showed that the karyotypes of Smilax and Heterosmilax are bimodal, with evidence of polyploidization, micro-variation, and variance of basic number from 16 to 15 and 13. We suggest that the karyotypes of the herbaceous Smilax species show a trend from symmetry in East Asian taxa to asymmetry in North America, but no such trend is obvious among woody species.

Regarding the inflorescence, we consider the prophyll to be a persistent bract. Therefore, the branches composed of single umbels borne in axils and with a prophyll at the base of the entire branch (i.e., sect. China and sect. Coilanthus) are homologous with the inflorescences possessing a prophyll at their base, or with a node on the peduncle and only a single umbel (in sect. Macranthae and sect. Smilax). Based on this interpretation and previous published ITS sequences, it is evident that a spike of umbels lacking peduncles is the primitive type of inflorescence in Smilacaceae from which two types of inflorescences are derived: (1) a solitary umbel without a prophyll, including species within Heterosmilax and Smilax section China and Coilanthus; and (2) a panicle-like type with one to many umbels subtended by a prophyll, i.e., the species of Smilax sect. Macranthae. Furthermore, our data suggest that Heterosmilax should not be recognized as a separate genus, but as a subgenus closely related to species of the herbaceous section of Smilax and Smilax sect. Coilanthus. The herbaceous species of Smilax, originating in East Asia and then expanding into North America, form a monophyletic group and a common ancestry with sect. Coilanthus. They are recognized as Smilax sect. Nemexia. Finally, studies of S. china show that it is a widespread species with varying ploidy levels. Nineteen leaf epidermis samples representing six species in Apios and two in Cochlianthus were examined for the first time using both light microscopy and scanning electron microscopy. Leaf epidermal characters of these two closely related genera, such as shape of cells, pattern of anticlinal walls, cuticular membrane and wax ornamentation, are usually stable within species and thus of great significance in understanding the relationships between and within genera. The results indicate that (1) A. gracillima Dunn should be recognized as a distinct species, not a variety of Apios delavayi Franch., (2) the division of Apios into two subgenera is unreasonable, and (3) Apios and Cochlianthus are retained as two distinct genera. Molecular data and new additional morphological data suggest that the two tree peonies, based on which Paeonia suffruticosa ssp. yinpingmudan was described as new, belong to two entities, the one from Anhui (K. Y. Pan and Z. W. Xie 9701, the type) being a member of P. ostii, while the other from Song Xian, Henan Province (D. Y. Hong et al. H97010) being a new taxon. P. suffruticosa ssp. yinpingmudan is here reduced as a synonym of P. ostii, and P. cathayana D. Y. Hong & K. Y. Pan is described as new based on the specimen from Henan. The new species is closely related to P. ostii and P. jishanensis, but it differs from the former in having lower leaves with 9 leaflets and petals rose, while from the latter in having leaflets glabrous, ovate or ovate-lanceolate, less lobed, sepals all caudate at the apex and petals rose. Anemone fulingensis W. T. Wang & Z. Y. Liu, a new species of the Ranunculaceae discovered from Fuling, Chongqing, China, is described and illustracted. In having sessile involucre bracts, 4-5-sepalled flowers, filiform filaments, and pantocolpate pollen it should be a member of Anemone sect. Stolonifera (Ulbr.) Juz., and from all other species of that section differs in its peculiar moniliform rhizome consisting of 2-10 tubers. In view of the fact that in Anemone the tubers are present in certain different evolutionary lines, and their appearance is a secondary phenomenon, this new species should be considered advanced in sect. Stolonifera.

Ranunculus ailaoshanicus W. T. Wang, a new species of the Ranunculaceae from Yunnan, China, is described and illustrated. This species in habit shows striking resemblance to Ranunculus dongrergensis Hand.-Mazz., and differs from the latter in its lower leafless scapes, smaller flowers, and smaller, oblong-elliptic petals with obscure nerves. In R. dongrergensis, the stems are 5–15 cm tall and bear cauline leaves, the flowers are larger, 1–1.8 cm in diam., and the petals are larger, obovate in outline, 4–9 mm long, 2.2–7 mm broad, and nervose. Roscoea cangshanensis M. H. Luo, X. F. Gao & H. H. Lin, a new species of the Zingiberaceae from Yunnan, China, is described and illustrated. Vanilla shenzhenica Z. J. Liu & S. C. Chen, a new species of Orchidaceae, is described and illustrated. This is the first new species of Orchidaceae found in Shenzhen, South China. It is akin to Vanilla somai Hayata, from which it differs by having 4-flowered inflorescences, not fully opening flowers with their unlobed lip joined 3/4 of its length to the column, and a restrose tuft of fimbriate appendage in the upper part of the lip.

A new species of the Ericaceae, Rhododendron tianmenshanense C. L. Peng & L. H. Yan, which was found to grow in forest in Zhangjiajie City, Hunan Province, China, was described and illustrated. The new species is similar to R. aganniphum Blaf. f. & K. Ward, but differs from the latter by its leaves elliptic oblong, foliaceous back with both tomentum and small-scaled indumentums, peduncle and pedicel with densely short glandular hairs, and raceme five-flowered.

A new species of the genus Drepanostachyum Keng f. of Bambusoideae from China, D. stoloniforme S. H. Chen & Z. Z. Wang, which was found to grow in Xiamen Botanical Garden, Fujian Province, is described and illustrated. A new liverwort species, Scapania paraphyllia T. Cao, C. Gao, J. Sun & B. R. Zuo, is described and illustrated from Zhejiang, China. The new species is closely related to S. bolanderi Aust. in having similar toothed leaves and pseudoparaphyllia. The former, however, differs from the later by having: (1) smaller plants; (2) pseudoparaphyllia numerous, not only growing on leaf axils but also on stems; (3) leaves divided into 1/4–1/3 of length and dorsal lobes as 1/3–1/2 size of ventral lobes; (4) cuticle of leaves rough, with large papillae up to 6–8 µm in diameter. The new species is also similar to S. ampliata Steph. with similar leaf shape and mouth of perianth. But it can be easily distinguished from the latter by having rough leaf cuticle and pseudoparaphyllia. Decontaminated thianthrene disproportion. Lagerstroemioxylon yuanmouensis sp. nov. is established from the Pliocene sediments near Xinhua, Yuanmou Basin, Yunnan, China. Its growth rings are distinct. Pores diffuse, solitary, and in radial multiples; perforation plates simple; intervessel pits alternate, vestured; vessel-ray pits alternate; tyloses thin-walled, abundant in vessels. Septate fibers distinct. Axial parenchyma predominantly paratracheal, narrow vasicentric and marginal. Rays mostly uniseriate, homocellular. Myrmechis bakhimensis D. Maity, N. Pradhan & G. G. Maiti (Orchidaceae), a new species from Sikkim Himalaya, is described and illustrated. The new species most closely resembles M. japonica (Reichb.) Gymnocoronis DC., a new naturalized genus of the tribe Eupatorieae, Asteraceae in China, was found in Guangxi, southern China. Morphologically it is related to Adenostemma, but differs in the absence of a pappus. Gymnocoronis spilanthoides (D. Don ex Hook. & Arn.) DC., the only one representative species of the genus in China, is described based on the specimens collected in Guangxi, China. Its distribution and possible pathways into China are also discussed.

Paphiopedilum helenae Aver., a newly recorded species of Orchidaceae from China, is reported. This species is characterized by having brightly yellow median sepals, orange ligulate or ligulate-spathulate petals with straight margin, brownish orange lip, and subcircular staminode. The populations are located on the karst cliffs near the peak of hills at altitude of 690 to 730 m. The plants grow under the evergreen broad-leaved forest. More than sixteen orchid species are found growing with P. helenae. The soil around the roots of the orchids was measured as pH 7.2. The growth of plants and the development of flower buds were positively correlated with the tree cover. Based on our two-year observation, however, we found very few flowers and fruits in the populations of this species, which might be most affected by resource limitation and pollinator limitation. At present the woodlands in this area were protected well, but the orchid populations still face potential threat because of their high commercial values. We, therefore, suggest that the conservation of this newly recorded orchid should be valued greatly and immediately.

Acer calcaratum and A. acuminatum, two poorly known and very rare species, occurring in Southeast Asia and the Himalayas respectively, were discovered recently in Yunnan Province and Xizang (Tibet) Autonomous Region in China. Acer calcaratum belongs to sect. Palmata ser. Sinensia, and is characterized by its 3 basally-nerved and 3-lobed leaves, with a rounded base, short, corymbose inflorescence and with few fruits, usually with only one large, well-developed samara, the nutlet more or less ovoid. Acer acuminatum belongs to sect. Arguta, and is characterized by its 3- or 5-lobed leaf blade, the lateral lobes usually as long as central lobe, apex of the lateral lobes caudate-acuminate, and infructescence 12–20 cm long. All were collected from Da’erbin Lake and marshes around it in the Da Hinggan Ling Mountains, Nei Mongol Autonomous Region, China. The type specimens of Aleuritopteris veitchii (Christ) Ching, A. duclouxii (Christ) Ching var. sulphurea Ching, and A. cremea Ching ex S. K. Wu were carefully examined. The lectotype of A. veitchii was designated based on the original description. A. cremea was identical to the above species and thus reduced to a synonym of it. It is characterized by concolorous, linear-lanceolate rhizome scales and finely dissected fronds with sulphur-yellow farina. A. duclouxii var. sulphurea, whose rhizome scales are distinctly bicolorous, can be easily distinguished from A. veitchii. The name of the above variety is reinstated. Based upon literature, examination of type specimens and field work, the distinguishing morphological characters among Carex fokienensis Dunn, C. minqingensis Z. P. Wang, C. jiuxianshanensis L. K. Dai & Y. Z. Huang and C. pallideviridis K. L. Chü (nom. nud.) were compared. From statistic analysis on perigynium length and comparison of shapes of pistillate scale, achene and perigynium, no prominent differences exist among C. fokienensis, C. minqingensis, C. jiuxianshanensis and C. pallideviridis, and these four taxa are taxonomically inseparable from each other. Thus, the later published C. minqingensis, C. jiuxianshanensis and C. pallideviridis should be reduced as synonyms of C. fokienensis. Decontaminated thianthrene disproportion. Based onherbarium and field observations and transplant experiment, Stemona sessilifolia (Miq.) Miq. was found to be a highly variable species in morphological characters such as habit, leaf shape, and the relative size of inner two perianth segments and the outer two perianth segments. In particular, the habit depends on whether the plant is growing in an open habitat, when it remains erect, or under forests when it is at first erect but then adopts a scandent habit. Thus it is clear that S. sessilifolia is a variable species and that S. shandongensis D. K. The correct name for arborescent lycopsid Lepidophloios Sternberg is discussed. Based upon Sternberg’s (1825) protologue, correlative literatures and type specimen, Lepidophloios is actually a later orthographical variant and its original spelling is Lepidofloyos. According to the International Code of Botanical Nomenclature (Vienna Code, Art. 60. 1 and 61. 1), Lepidofloyos should be the correct name for Lepidophloios.
